Molecules That Affect Megakaryocyte Development

چکیده

RESEARCH QUESTION Research into megakaryocyte activity has picked up in recent years, largely due to the introduction of more refined methods of studying these large multinucleated cells. More studies are being conducted on various molecules that may interact with and control megakaryocyte development, and subsequently their end-product, the platelets. The enzyme acetylcholinesterase (AchE) has been found to associate with megakaryocytes during their growth and development (Powers, 1989; Straneva et al, 1986). Another molecule, thrombopoietin [TPO], is also known to influence megakaryocyte development (Powers, 1989; Straneva et al, 1986). The focus of my research will be to investigate the effect acetylcholinesterase has on megakaryocyte biology in the presence and absence of thrombopoietin. My hypothesis is that acetylcholinesterase expression is enhanced in the presence of thrombopoietin, and this in turn influences the growth and development of megakaryocytes.

Two ellagic acids isolated from roots of Sanguisorba officinalis L. promote hematopoietic progenitor cell proliferation and megakaryocyte differentiation.

Using a bioassay-directed chromatographic separation, two ellagic acids were obtained from the ethyl acetate extract of the roots of Sanguisorba officinalis L. On the basis of chemical and spectroscopic methods, the two ellagic acids were identified as 3,3',4-tri-O-methylellagic acid-4'-O-β-d-xyloside and 3,3',4-tri-O-methylellagic acid. Stimulation of cell proliferation was assayed in hematopo...

Recombinant human megakaryocyte growth and development factor stimulates thrombocytopoiesis in normal nonhuman primates.

Megakaryocyte growth and development factor (MGDF) is a novel cytokine that binds to the c-mpl receptor and stimulates megakaryocyte development in vitro and in vivo. This report describes the ability of recombinant human (r-Hu) MGDF to affect megakaryocytopoiesis in normal nonhuman primates. r-HuMGDF was administered subcutaneously to normal, male rhesus monkeys once per day for 10 consecutive...

Evaluating the Prognostic Significance of Bone Marrow Megakaryocyte Count in Patients with Idiopathic Thrombocytopenic Purpura

Background: Previous studies about the relationship between bone marrow megakaryocyte count and the chronicity of ITP has yielded paradoxical results. The aim of the present study was to investigate any relationship between the megakaryocyte count in the bone marrow and the chronicity of ITP. Materials and Methods: This study was performed to compare the primary bone marrow aspiration megakar...

High-content live-cell imaging assay used to establish mechanism of trastuzumab emtansine (T-DM1)--mediated inhibition of platelet production.

Proplatelet production represents a terminal stage of megakaryocyte development during which long, branching processes composed of platelet-sized swellings are extended and released into the surrounding culture. Whereas the cytoskeletal mechanics driving these transformations have been the focus of many studies, significant limitations in our ability to quantify the rate and extent of proplatel...

GATA1-mediated megakaryocyte differentiation and growth control can be uncoupled and mapped to different domains in GATA1.

The DNA-binding hemopoietic zinc finger transcription factor GATA1 promotes terminal megakaryocyte differentiation and restrains abnormal immature megakaryocyte expansion. How GATA1 coordinates these fundamental processes is unclear. Previous studies of synthetic and naturally occurring mutant GATA1 molecules demonstrate that DNA-binding and interaction with the essential GATA1 cofactor FOG-1 (...
